The goal of this project is to determine which residues within 15-LOX-2, a human lipoxygenase enzyme, are important for the interactions with the allosteric effector, oleyl sulfate (OS), which has been shown in the Offenbacher lab to alter the substrate preference and rate limiting steps of catalysis for 15-LOX-2. Evidence from recent models, developed by Dr. Yumin Li's lab, supports the role of three amino acid residues (with His627 being the primary target residue) in the PLAT regulatory domain.
