Objectives: A. Refine the thermal network of LOX enzyme as a model tunneling system. Validate the thermal network by mutagenesis studies with kinetic assays. Explore the use of selective ?thermal? pumps to accelerate catalysis from the refined thermal network. B. Collect and analyze HDX-MS and map the thermal network(s) in YADH C. Guided design and/or development of tools to enhance tunneling (cavity) D. Design and investigate ancestral enzymes to understand and promote tunneling with evolution
