abstract

  • Pyruvate carboxylase (PC) activity that is not carefully regulated contributes to the progression of type 2 diabetes and cancer. The complex mechanism which is responsible for PC regulation is unknown. We propose to develop two novel techniques in our lab that will allow us to accomplish our long-term research goal of determining the regulatory mechanism of PC at the molecular level. Completion of our research will aid in (1) defining the biochemical basis for the persistent gluconeogenesis characteristic of type 2 diabetics, (2) rationalizing the observed increase in PC activity in certain tumor cells and (3) developing more effective mechanistic-based therapeutics.
